Effects of Adrenergic Drugs on Blood Concentrations of Insulin and Cyclic AMP in Streptozotoc n-induced Diabetic Rats


Abstract
This study was conducted to ellucidate the effects of adrenergic drugs on the blood concentrations of insulin and cyclic AMP in streptozotocin-induced diabetic rats. The results obtained were as follows.
1. Hyperglycemia in streptozotocin-induced diabetic rats was potentiated by isoproterenol and phentolamine to 146% and 174% of the basal level respec-tively, but suppressed by propranolol and oxprenolol to 77% and 75% of the basal respectively and not significantly changed by tolazoline and phenoxyben-zamine.
2. Blood insulin concentrations in streptozotocin induced diabetic rats were decreas-ed by combined therapy of propranolol and phentolamine to 83% of those in phentolamine alone therapy and not significantly changed by other drugs.
3. Blood cAMP concentrations in streptozotocin-induced diabetic rats were increas-ed. by isoproterenol and phentolamine to 3.9 and 4.1 times of the basal level respectively, decreased by combined therapy of propranolol and phentolamine to 89% of those in phentolamine alone therapy and not significantly changed by other drugs.
These results indicated that isoproterenol and phentolamine potentiated the hyperglycemia in streptozotocin-induced diabetic rats through increasing blood cAMP concentration and propranolol and oxprenolol suppressed without changes in blood cAMP concentration.
Therefore the symptoms of diabetes mellitus could be deteriorated by administra-tion of isoproterenol, phentolamine and other drugs which induce the increase in blood cAMP concentration.
